The mechanism of amine inhibition of the photosynthetic oxygen evolving complex Amines displace functional chloride from a ligand site on manganese

چکیده
Amines inhibit photosynthetic oxygen evolution by binding to manganese in the oxygen evolving complex. This inhibition is more effective if chloride is absent. Steady-state kinetic analyses show that ammonia and chloride compete for the same site in the oxygen evolving complex; inhibition of oxygen evolution by either chloride depletion or by ammonia addition induces Signal IIr, which saturates at high microwave power. These data indicate that chloride is bound at the manganese site in the oxygen evolving complex. We propose that the anion functions as a bridging ligand mediating electron transfer between manganese atoms and perhaps also between manganese and Z, the primary donor to P680.

Amine binding and oxidation at the catalytic site for photosynthetic water oxidation.
Photosynthetic water oxidation occurs at the Mn-containing catalytic site of photosystem II (PSII). By the use of 14C-labeled amines and SDS-denaturing PAGE, covalent adducts derived from primary amines and the PSII subunits, CP47, D2/D1, and the Mn-stabilizing protein, can be observed. When PSII contains the 18- and 24-kDa extrinsic proteins, which restrict access to the active site, no 14C la...

A tetra-manganese cluster in the photosystem II (PSII) pigment-protein complex plays a critical role in the photosynthetic oxygen evolution process. PsbY, a small membrane-spanning polypeptide, has recently been suggested to provide a ligand for manganese in PSII (A.E. Gau, H.H. Thole, A. Sokolenko, L. Altschmied, R.G. Herrmann, E.K. Pistorius [1998] Mol Gen Genet 260: 56-68). We have construct...

A new two-dimensional manganese(II) coordination polymer, [Mn(μ-DEP)(μ-Cl)(H2O)2]n (1) (DEP is diethyl phosphate), was prepared from the reaction of MnCl2.4H2O and TEP (TEP is triethyl phosphate) ligand in mixture of H2O/CH3OH (1:10) at 40 °C. Suitable crystals of this complex for crystal structure determination were obtained by slow evaporation of the produced colorless solution at room temper...

Macrocyclic heterobinuclear Zn(II)–Cu(II) complexes with phenol based dicompartmental ligands possessing contiguous hexa- and penta-coordination sites were prepared by a stepwise procedure. The ligands include similar N4O2 and dissimilar N(imine)3O2 and N(amine)3O2 coordination sites sharing two phenolic oxygen atoms. The six-coordination site comprises two pyridyl pendant arms on the amine nit...
متن کاملInhibition of chickpea seedling copper amine oxidases by tetraethylenepentamine
Copper amine oxidases are important enzymes, which contribute to the regulation of mono- and polyamine levels. Each monomer contains one Cu(II) ion and 2,4,5-trihydroxyphenylalanine (TPQ) as cofactors. They catalyze the oxidative deamination of primary amines to aldehydes with a ping-pong mechanism consisting of a transamination. The mechanism is followed by the transfer of two electrons to mol...
